Talleres were impressive in the group stage of this year's competition, boasting a record of three wins, two draws and one defeat from their six matches, which saw them claim 11 points to finish second in Group H.
The Blue and Whites play their league football in the Argentine Primera Division, finishing third in the table last term, picking up 46 points from their 25 matches.
Talleres have made a disappointing start to their new league campaign, though, only collecting four points from five matches, which has left them down in 24th spot in the table.
Pedro Caixinha's side suffered a 2-0 defeat to Central Cordoba SdE in their last league match on June 25, but they are unbeaten in their last four matches in this competition, winning their final group game against Universidad Catolica at the end of May.
Colon, meanwhile, also play their league football in Argentine's top flight, finishing seventh in the 2021 season, seven points behind their opponents in this two-legged Copa Libertadores last-16 affair.
Julio Cesar Falcioni's side picked up 10 points from their six Group G fixtures in this competition to finish at the head of the section, two points ahead of second-place Cerro Porteno.
Colon have struggled in the early stages of their 2022 league season, though, collecting just five points from five matches to sit 20th in the table.
The Santa Fe-based outfit have actually lost their last three matches in all competitions, with their last success coming against River Plate in the league in the middle of the month.
Colon will have home advantage in the second leg of this last-16 tie on July 6, so it will be interesting to see how head coach Falcioni approaches Tuesday's contest.

Talleres had Christian Oliva sent off in their league defeat to Central Cordoba SdE last time out, but the midfielder is available for this contest and is expected to start.
The home side have no injury concerns heading into the match, but there could be a couple of changes from the side that started their league game on Saturday.
Enzo Diaz is pushing to return to the XI, while there could also be a spot in the side for Diego Ortegoza.
As for Colon, Augusto Schott and Andrew Teuten will miss the contest through injury, but the visitors are otherwise in strong shape heading into the first leg of their last-16 affair.
Facundo Hernan Farias is again expected to lead the line for Falcioni's side, with Santiago Pierotti likely to play off the right on Tuesday.
There could be a change in the middle of the park, though, with Leandro Ojeda bidding to replace Tomas Moschion, who was replaced at the interval against Huracan in the league on Saturday.
