The former Chelsea, Real Madrid, Manchester United and Tottenham manager could not contain his delight when Stephan El Shaarawy put the ball into the back of the net in the 92nd minute against Sassuolo.
Mourinho took off running down the touchline, in classic Jose style, to celebrate with his players.
He jumped on and hugged Tammy Abraham as he got to the Roma stars in the brilliant scenes at the Stadio Olympico.
The 58-year-old clearly has not lost his love of the game and it was brilliant to see another classic celebration from the manager in this huge landmark match.
It was also very reminiscent to the time he ran down the touchline after Porto had netted a decisive injury-time goal against Manchester United in the Champions League back in 2004.
Speaking after the game, Mourinho said: "I told everyone lies! I had been saying to the team for days that this 1000th game was not important to me… but I had an incredible fear of losing!
"Luckily we won and I was running like a child today because I was feeling 12 years old, not 58."
Mourinho has now racked up 1000 games in management, winning an incredible 639 of them and with 25 trophies to his name.
His managerial career has seen him coach in four different countries with Porto, Chelsea, Inter Milan, Real Madrid, Chelsea [again], Manchester United, Tottenham and now Roma.
