The 21-year-old made his first appearance for Los Blancos off the bench against Osasuna before the November international break, with the centre-back required after Eder Militao picked up a season-ending knee injury.
Lucas Vazquez was also injured in that match, so there will be two changes at the back, with Federico Valverde expected to be asked to operate as the right-back, as Dani Carvajal is also out for the season with an ACL issue.
Rodrygo was the third Real Madrid player to pick up an injury in the 4-0 victory over Osasuna, and his absence is expected to open the door for Brahim Diaz to feature in the final third of the field.
Luka Modric will feature alongside Jude Bellingham and Eduardo Camavinga in midfield, with Vinicius Junior and Kylian Mbappe both starting in the final third of the field.
There should also be a huge injury boost for Real Madrid, with Thibaut Courtois back in training, so the Belgian is in line to replace Andriy Lunin after recently being sidelined with a muscular problem.
As well as Militao, Vazquez, Carvajal and Rodrygo, David Alaba is still out, with the Austrian not expected back until January, while Aurelien Tchouameni will be missing until the start of December due to an ankle problem.
Real Madrid possible starting lineup: Courtois; Valverde, Asencio, Rudiger, Mendy; Modric, Camavinga, Bellingham; Diaz, Mbappe, Vinicius
