The home team will enter the contest off the back of a goalless draw with Barrow on Saturday, while Grimsby recorded a 1-0 victory over Rochdale on the same afternoon.
Match preview
Sutton impressed in their first ever campaign in the Football League last season, finishing eighth in the division, only one point outside of the playoff positions.
The Yellows are again in the playoff picture at this stage, currently sitting ninth in the table, boasting 55 points from their 36 matches, just one point behind seventh-placed Salford City.
Sutton are actually unbeaten in the fourth tier of English football since the middle of January, winning five and drawing four of their last nine matches at this level of football.
Matt Gray's side have been excellent at home this season, picking up 37 points from 18 matches in front of their own supporters, but Grimsby have won seven times on their travels this term and are certainly capable of claiming a positive result against their in-form hosts.
The two teams played out a goalless draw in the reverse match earlier this season, but Sutton will undoubtedly be looking for the win here to boost their playoff hopes.
Grimsby, meanwhile, will enter this match off the back of a 1-0 victory over Rochdale on Saturday, with George Lloyd scoring the only goal of the contest in the 68th minute.
A record of 11 wins, nine draws and 13 defeats has seen the Mariners collect 42 points, which has left them in 16th position in the table, 13 points outside of the relegation zone and 14 points from the top six.
Grimsby have only lost one of their last seven matches in all competitions, which includes their famous 2-1 victory over Southampton in the last round of the FA Cup.
Paul Hurst's side will face Brighton & Hove Albion in the quarter-finals of the competition next weekend, and it would be some story if they were able to progress to the semi-finals of the tournament.
Grimsby were promoted back to League Two via the National League playoffs this season, so consolidating back at this level is the team's main aim for the next couple of campaigns.

Team News
Sutton have no fresh injury concerns from their goalless draw at Barrow on Saturday afternoon.
As a result, it could be a case of same again in terms of the starting side, with Lee Angol and Omar Bugiel operating in the final third of the field.
Matthew Dennis and William Randall-Hurren both featured off the bench last time out and are options to come into the XI here, but head coach Gray could decide to keep faith with the same side from Saturday.
Grimsby, meanwhile, will need to make a check on the fitness of James McAtee, who was replaced at the interval of the league win over Rochdale on Saturday afternoon.
Should McAtee prove his fitness, then head coach Hurst could name an unchanged side from the one-goal success last time out.
Lloyd will again lead the line for the away team, while it is set to be the same formation, with a back three including the experienced Luke Waterfall.
Sutton United possible starting lineup:
Rose; Kizzi, Rowe, Goodliffe, Hart; Ajiboye, Eastmond, Smith, Boldewijn; Angol, Bugiel
Grimsby Town possible starting lineup:
Crocombe; Efete, Waterfall, Maher; Emmanuel, Green, Holohan, Driscoll-Glennon; McAtee, Lloyd, Khan
We say: Sutton United 1-1 Grimsby Town
Sutton's home form has been excellent this season, but Grimsby have also won seven times on their travels, so we are expecting a close match on Tuesday night. Grimsby have shown that they are capable of picking up some big results this season, so we are tipping the Mariners to avoid defeat here.
